Synthesize a baculovirus-optimized shuttle vector for your desired protein, generate high purity baculovirus, amplify virus to 250 ml high titer stock and determine the titer by limiting dilution.
The transfer vector you provide will be used to develop a high purity virus and the virus amplified to 250 ml high titer virus, and the titer determined.
Your virus will be cloned by limiting dilution on 96 well dishes. 5 clones will be used to test infect one T-25 flask each and the pellet (and media if appropriate) will be shipped to you for analysis. The chosen clone will be amplified to 250ml and the titer will be determined.
Your clonal pellets will be applied to an SDS-PAGE gel, blotted and probed with an antibody you provide or using any commercially available antibody. Detection by ECL. Maximum 14 clones including controls per gel.
Your cells will be infected by virus at an MOI >1, or as you direct, and harvested 48 hours later. Volume discounts 2-4L 20%, 5-9L 33.3%, 10L or more, 50%.
Your protein will be solubilized and purified by one column using an affinity method of your choice.
Cells expressing your protein and appropriate controls will be fractionated into cytoplasmic, detergent-soluble and detergent-insolouble fractions, analyzed by gel electrophoresis, blotted and probed with an antibody you provide or using any commercially available antibody. Detection by ECL.
